P 416 Lipofuscin accumulation in the retinal pigment epithelium and fatty acid diet

چکیده
B Confirming the diagnosis of night blindness in our patient, the scotopic response in his electroretinogram (ERG) was distinctly reduced while the photopic response was normal. When his blood was examined for a low level of vitamin A as a possible cause for night blindness, we found the value to be reduced below normal (17 pg/dl versus 25-70 pg/dl in normal subjects). Treatment with vitamin A supplement resulted in prompt normalization of vitamin A level (79 pg/dl after two weeks). Symptoms completely disappeared, and the scotopic response in the ERG rehuned to normal.

Ccl2/Cx3cr1-deficient mice: an animal model for age-related macular degeneration.
BACKGROUND/AIMS Senescent Ccl2-/- mice develop cardinal features of human age-related macular degeneration (AMD). Loss-of-function single-nucleotide polymorphisms within CX3CR1 are associated with AMD. METHODS We generated Ccl2-/-/Cx3cr1-/- [double-knockout (DKO)] mice and evaluated the eyes using fundoscopy routine histology, immunochemistry, biochemistry and proteomics. RESULTS At 6 weeks...
متن کاملAccelerated accumulation of lipofuscin pigments in the RPE of a mouse model for ABCA4-mediated retinal dystrophies following Vitamin A supplementation.
PURPOSE Dietary supplementation with vitamin A is sometimes prescribed as a treatment for retinitis pigmentosa, a group of inherited retinal degenerations that cause progressive blindness. Loss-of-function mutations in the ABCA4 gene are responsible for a subset of recessive retinitis pigmentosa. Other mutant alleles of ABCA4 cause the related diseases, recessive cone-rod dystrophy, and recessi...
متن کاملIron-induced accumulation of lipofuscin-like fluorescent pigment in the retinal pigment epithelium.
PURPOSE One of the most prominent changes that occurs in the retinal pigment epithelium during senescence is the progressive accumulation of the autofluorescent pigment lipofuscin. Experiments were conducted to evaluate the role of nonenzymatic oxidation of photoreceptor outer segments in retinal pigment epithelium lipofuscin formation. METHODS Albino Fischer rats were given intravitreal inje...
متن کاملReduced phagosomal content of the retinal pigment epithelium in response to retinoid deprivation.
Previous investigations have shown that lipofuscin accumulation in the retinal pigment epithelium (RPE) is reduced greatly as a consequence of vitamin A deprivation. The mechanism by which vitamin A regulates RPE lipofuscin deposition remains to be determined. It is possible that retinoids are direct precursors of this substance. Alternatively, vitamin A deficiency may reduce the uptake and pro...
متن کاملLipids in human lipofuscin-enriched subcellular fractions of two age populations. Comparison with rod outer segments and neural retina.
The fatty acid composition and content of total phospholipids, free fatty acids (FFA), diacylglycerols (DG), phosphatidylcholine (PC), phosphatidylserine (PS), and phosphatidylethanolamine (PE) were studied in lipofuscin granules of human donors in two age groups, young (less than 40 yr old) and old (more than 47 yr old), and compared with lipids of the photoreceptor rod outer segments (ROS). N...
